Understand the connection

COMT helps process dopamine, norepinephrine, epinephrine, and catechol estrogens using methyl groups from SAM. Its activity depends on more than one common variant.

Follow the pathway

Dr. Purser discusses rs4680 and rs4633 alongside regulatory and linked variants such as rs6269, rs4818, rs4646312, and rs165722. He connects COMT with stress response, pain sensitivity, sleep, and estrogen processing. Linked groups of variants are called haplotypes; a label such as “slow COMT” is a simplification, not a psychiatric diagnosis.

Bring these questions to your visit

Ask which exact variants and alleles are present and whether the interpretation considers them together. Review magnesium, methylation-related nutrients, stimulant exposure, and medicines with your clinician before changing them. A homozygous wild-type result is different from a homozygous risk allele.
